Mécanismes d’action
- principal
*Action antiinflammatoire au stade aigu de l’inflammation :
– inhibe l’augmentation de la permabilit capillaire,
– diminue la migration des polynuclaires des monohistiomacrophages,
– inhibe la sortie des enzymes lysosomiaux,
– s’oppose l’action des mdiateurs chimiques,
– inhibe de faon non spcifique la synthse des prostaglandines,
– stimule la scrtion de l’ACTH.
Aucune action sur les phnomnes tardifs de l’inflammation : en particulier, n’empche pas la formation du tissu de granulation.
*Action analgsique priphrique : type aspirine mais moins actif.
*Action antipyrtique type aspirine qui serait due l’inhibition des prostaglandines synthtases notamment PGE1. - secondaire
Inducteur enzymatique des enzymes microsomiaux hpatiques qui assurent la biotransformation des mdicaments.
Rtention hydrosode par action directe sur les tubules rnaux pouvant augmenter le volume plasmatique jusqu’ 50%.

- TOXICITE FOETALE
Au cours du troisime trimestre de grossesse, tous les inhibiteurs de synthse des prostaglandines peuvent exposer le foetus une toxicit cardiopulmonaire (hypertension pulmonaire avec fermeture prmature du canal artriel).
